I have purchased a Marriott timeshare property from the developer and another on the resale market. The only thing you lose by buying on the resale market is the ability to trade your unit for Marriott points. For example, I own a Marriott floating week at Hilton Head that can be traded for 110,000 Marriott points. These points can be used at Marriott hotels and resorts world wide. This is not a big incentive, in my opinion, for buying from the developer when one considers the considerable discount one receives when buying resale. A third option you did not mention is buying resale though Marriott. The third option gives you some discount along with the Marriott points.
You can join both RCI and II when buying a Marriott resale. II is preferable, though, if you are buying a Marriott.
Your last question concerned whether the seller of a Marriott timeshare can transfer Marriott points to the buyer. The answer is no.
